Sinequan belongs to a drug class known as tricyclic antidepressants (TCAs). The main function of these drugs is to treat symptoms of depression and anxiety. Depression is usually caused by an imbalance in the chemicals of the brain, known as neurotransmitters. Sinequan, under the generic name Doxepin hydrochloride, targets the neurotransmitters serotonin, norepinephrine, acetylcholine, and histamine. Although the exact mechanism of action is unknown, it is theorized that Sinequan elevates mood in depression by increasing the levels of available serotonin and norepinephrine, and blocks the activities of acetylcholine and histamine. It can also be used to treat depression caused by alcoholism, or brain damage, or depression caused by bipolar disorder.
Sinequan is available in 10, 25, 50, 75, 100, and 150 mg capsules. It is also available in oral concentrates of 10 mg/mL. The usual dose is from 75 to 300 mg daily. It can be administered once at bed time, or in 3 divided doses. More than the 300 mg limit is not recommended and bring unwanted side effects. It cannot be used with drugs such as monoamine oxidase inhibitors and cimetidine. MAOIs such as Parnate can cause high fever and convulsions when used concurrently with TCAs.
