How does Saizen work? What will it do for me?

Saizen belongs to a class of drugs known as growth stimulants. Saizen is a man-made version of growth hormone. Growth hormone is made naturally in our bodies by the pituitary gland and is necessary for stimulation of growth in children.

How should I use Saizen?

Saizen is administered by injection only. It is usually injected subcutaneously (under the skin), although it can be given intramuscularly (into the muscle) if the doctor prefers. If it is injected under the skin, it is important to rotate the site of injection in order to minimize the risk of fat wasting at the site of injection.

The dose depends on the circumstances and body weight of the person being treated. The recommended starting dose is 0.2 mg per kilogram of body weight per week. The maximum dose is 0.27 mg per kilogram per week. The weekly dose should be divided into equal doses and given either on three alternate days, six times per week, or daily.

If you are using this medicine at home, your doctor will ensure that you understand exactly how to use this medicine. Make sure you ask any and all questions to ensure that you use the medicine exactly as prescribed. Please refer to the appropriate package insert for details on how to mix the medicine and for proper injection procedures. It is important to visit your doctor regularly during therapy to ensure the Saizen kit is working safely and effectively.

If you miss a dose of the medicine, take it as soon as you remember. If it is the next day before you remember your missed dose, call the doctor for further instructions. Do not double-up doses.

Many things can affect the dose and schedule of medication that a person needs, such as body weight, other medical conditions, and other medications. Your doctor may recommend a dose or schedule different from the ones listed here.

Saizen comes as vials of powder that are mixed with the liquid provided (called a diluent) to create a solution. The 1.33 mg and 3.33 mg vials should be used immediately after they are mixed, provided that they are mixed with the diluent provided in the package (Sodium Chloride). The 3.33 mg vial may also be mixed with a different diluent called Bacteriostatic Sodium Chloride. In this case, it may be stored in the refrigerator for up to 14 days after mixing. The 5 mg vials may be stored in the refrigerator for up to 14 days after mixing with the diluent provided in the package (Bacteriostatic Water for Injection). If the 5 mg vials are mixed with a different diluent called Water for Injection, they should be used immediately. Do not use the solutions if they are discoloured or contain particles.

Saizen injection should not be used by:

anyone allergic to any of the ingredients in the product being used
children who have closed epiphyses
anyone with active cancer
anyone with a brain tumour


What side effects are possible with Saizen?

The following side effects may go away as your body becomes used to the medicine; check with your doctor if they continue or become bothersome.

Less common or rare (Less than 10%)

carpal tunnel syndrome
growth of birthmarks
joint pain
muscle pain
swelling of hands, feet, or lower legs
unusual tiredness or weakness
Contact your doctor as soon as possible if any of the following occur:

ear infection or other ear problems (in Turner's syndrome)

abdominal pain or bloating
depression of skin at place of injection
headache
limp
nausea and vomiting
pain and swelling at site of injection
pain in hip or knee

Are there any other precautions or warnings for Saizen?

Diabetes: For people with diabetes, the dose of insulin might require adjustment with Saizen is started. Blood sugars should be closely monitored and necessary adjustment in insulin therapy should be made with a doctor's advice.

Hypothyroidism (low level of thyroid hormone): People with untreated hypothyroidism will have an inadequate response to Saizen therapy. They should be monitored while taking Saizen to ensure that they are getting the proper dose.

Scoliosis: People with scoliosis are at risk of a worsening of their condition while taking Saizen; therefore, these patients should be cautious.

Pregnancy: It is not known whether Saizen is harmful when used during pregnancy. It should be used during pregnancy only if potential benefits outweigh risks.

Breast-feeding: It is not known whether Saizen is excreted in breast milk. It should be used with caution by nursing women.

What other drugs could interact with Saizen?

The following agents may affect how Saizen works, or increase the occurrence of side effects:
corticosteroids (e.g., prednisone)
cyclosporine
estrogen
testosterone
If you are taking any of these medications, speak with your doctor or pharmacist. Depending on your specific circumstances, your doctor may want you to:

stop taking one of the medications,
change one of the medications to another,
change how you are taking one or both of the medications, or
leave everything as is.
An interaction between two medications does not always mean that you must stop taking one of them. In many cases, interactions are intended or are managed by close monitoring. Speak to your doctor about how any drug interactions are being managed or should be managed.

Medications other than those listed above may interact with Saizen. Tell your doctor or prescriber about all prescription, over-the-counter (non-prescription), and herbal medications that you are taking. Also tell them about any supplements you take. Since caffeine, decongestants, alcohol, the nicotine from cigarettes, or street drugs can affect the action of many medications, you should let your prescriber know if you use them.
