Manufactured by Wyeth-Ayerst Laboratories. Synvisc® (Hylan G-F 20) is an FDA-approved therapy that provides lubrication for your knee. It is a viscosupplement used to treat the pain of osteoarthritis of the knee. It also acts as a "shock absorber" to cushion your knee joint. Synvisc® is an elastic and viscous fluid that is made from a substance called hyaluronan that is found in normal joint fluid. Hyaluronan acts as a "shock absorber" and lubricant in your knee joint and is needed for your joint to work properly.

Synvisc Caution
Synvisc is generally well tolerated; however, it may not work for everyone. The most commonly reported side effects are temporary pain, swelling, and/or fluid accumulation in the injected knee (about 2% of injections). If fluid accumulation is large or painful, call your doctor. Other side effects, such as rash, have been reported rarely. Before you begin your treatment with Synvisc®, advise your doctor if you are allergic to products from birds, such as feathers, eggs, or poultry, or your leg is swollen or infected. The results of repeat use have not been established. FOR WOMEN: If you plan on becoming pregnant, discuss with your doctor the benefits and risks of using this medicine during pregnancy. It is unknown if this medicine is excreted in breast milk. If you are or will be breast-feeding while you are using this medicine, check with your doctor or pharmacist to discuss the risks to your baby.

What is Synvisc Hylan?

Hylan G-F 20 is a fluid containing hylan polymers produced from chicken combs.

How does Synvisc Hylan work?

The exact mechanism is unknown, however it is speculated that the quality and quantity of hyaluronate in the joint fluid and tissues may be deficient in those with OA. Adding Hylan G-f 20 allows the joint to function better.

Synvisc Hylan Dosage: Hylan G-f 20 is supplied in ml pre-filled syringes. It is administered by intra-articular injection once a week (1 week apart), for a total of 3 injections. 

How should I take Synvisc Hylan?

Your doctor will inject your knee once a week for 3 weeks.

Synvisc Contraindications: Those allergic to products from birds such as feathers, eggs, and poultry should avoid this product.

The safety and effectiveness of Hylan G-f 20 has not been studied in children. 

Sodium Hylan G-f 20 should be avoided by those with known hypersensitivity to hyaluronate preparations.

Intra-articular injections are contraindicated in cases of infections or skin diseases in the area of the injection site.

The safety and effectiveness of the use of Sodium Hylan G-f 20 in joints other than the knee have not been established.

The safety and effectiveness of sodium Sodium Hylan G-f 20 have not been established in pregnant or nursing women.

Should be used with caution when there is evidence of lymphatic or venous stasis in that leg.

Synvisc Hylan Interactions:

Synvisc Should not be used with disinfectants containing quatemary ammonium salts for skin preparation because hyaluronic acid can precipitate in their presence.

Do not inject anesthetics or other medications into the knee joint during Synvisc therapy. Such medications may dilute Synvisc and affect its safety and effectiveness.

Possible Synvisc Hylan side effects:

These side effects are not considered serious but are certainly annoying for those experiencing them. The most frequently reported side effects include gastrointestinal complaints, headache, injection site pain, knee swelling/effusion, and rash, bruising, and itching at the injection site.

Signs of infection at the injection site include fever, redness, swelling and pain. If you suspect an infection notify your doctor right away.

Signs of a possible allergic reaction are rash, swelling and difficulty breathing. These symptoms should be reported immediately. 

SYNVISC DESCRIPTION

Synvisc (hylan G-F 20) is an elastoviscous fluid containing hylan polymers produced from chicken combs. Hylans are derivatives of hyaluronan (sodium hyaluronate), a natural complex sugar of the glycosaminoglycan family. Hyaluronan is a long-chain polymer containing repeating disaccharide units of Na-glucuronate-N-acetylglucosamine.

 

SYNVISC WARNINGS

Do not concomitantly use disinfectants containing quaternary ammonium salts for skin preparation because hyaluronan can precipitate in their presence.

Do not inject Synvisc extra-articularly or into the synovial tissues and capsule. Local and systemic adverse events, generally in the area of the injection, have occurred following extra-articular injection of Synvisc.

Intravascular injections of Synvisc may cause systemic adverse events.

 

 

SYNVISC DIRECTIONS FOR USE

Synvisc is administered by intra-articular injection once a week (one week apart) for a total of three injections.

Precaution: Do not use Synvisc if the package has been opened or damaged. Store in original packaging (protected from light) at room temperature below 86°F (30°C). DO NOT FREEZE.

Precaution: Strict aseptic administration technique must be followed.

Precaution: Do not concomitantly use disinfectants containing quaternary ammonium salts for skin preparation because hyaluronan can precipitate in their presence.

Precaution: Remove synovial fluid or effusion before each Synvisc injection.

Do not use the same syringe for removing synovial fluid and for injecting Synvisc, but the same needle should be used.

Take particular care to remove the tip cap of the syringe and needle aseptically.

Inject Synvisc into the knee joint through an 18 to 22 gauge needle.

To ensure a tight seal and prevent leakage during administration, secure the needle tightly while firmly holding the luer hub.

Precaution: Do not over tighten or apply excessive leverage when attaching the needle or removing the needle guard, as this may break the tip of the syringe.

Do not inject anesthetics or any other medications intra-articularly into the knee while administering Synvisc therapy. This may dilute Synvisc and affect its safety and effectiveness.

Precaution: The syringe containing Synvisc is intended for single use. The contents of the syringe must be used immediately after the syringe has been removed from its packaging. Inject the full 2 mL in one knee only. If treatment is bilateral, a separate syringe must be used for each knee. Discard any unused Synvisc.
